  • Open Pulled Straw
Can I use the solution if it has frozen?

If the solution freezes, we recommend that you discard it immediately. As stated in the User Guide, the product should be stored at a temperature of between 2–8°C. When stored unopened within this range, the product will remain stable and ready to use until the date shown on the bottle. Media should not be exposed to temperatures outside this range at any time.

Can embryos cultured in culture media from other manufacturers be vitrified or warmed with VitaVitro kits?

Yes. VitaVitro vitrification and warming solutions can be used to vitrify or warm embryos that have been cultured in any CE- or FDA-approved embryo culture media.

How long does the product last after the bottle has been opened?

Once opened, it is recommended to use the solution within one day, assuming it is used under aseptic conditions in closed vials. Discard unused residual solution.

How many vitrification and warming cycles can be carried out with one vitrification & warming kit?

If used in accordance with the User Guide, one kit can be used for 5 vitrification cycles and 1–2 warming cycles.

How many samples can be cryopreserved in one cycle and with one VitaVitro kit?

Under normal circumstances:

  • 1–3 oocytes/embryos can be cryopreserved in one cycle.
  • 10–15 oocytes/embryos can be cryopreserved with one VitaVitro kit.
  • The number of samples may vary depending on the cryo-device and the protocol used in your laboratory.

Do VitaVitro vitrification & warming solutions contain antibiotics?

No. VitaVitro vitrification and warming media do not contain antibiotics due to the increased risk of intracellular accumulation and toxic damage during the cryopreservation process. The process of equilibration and dilution is relatively short, and all media used before and after cryopreservation should contain antibiotics. As stated in the User Guide, vitrification and warming media, like all other aspects of the procedure, should be handled using aseptic technique. Based on extensive experience worldwide, these measures are sufficient to prevent infections.

What is the appropriate volume of vitrification solution in the OPS after loading?

We recommend approximately 1 µl of vitrification solution for sample loading. Using an insufficient amount of solution may affect the capillary suction and the formation of a solution column at the end of the straw, while using too much solution may compromise the cooling and warming rate.

How many samples can be cryopreserved in a single OPS?

A maximum of 3 samples in one straw is recommended.

What should I do if vitrification solution remains in the straw after warming?

The recommended solution is to insert a 20 μl micropipette with a narrow plastic tip into the wide end of the OPS and gently expel air into the OPS to eject the solution column into the warming dish. To avoid delays, we suggest having a pipette and tip ready for use before starting the procedure.

What kind of sealer should I use?

Sealing must be performed with a handheld heat sealer adjusted to 100–120°C. Note: Approximately 70% of the container straw should be kept immersed in LN2 during the sealing process. VitaVitro offers a purpose-designed handheld sealer for this procedure.
